ICS 65.020 CCS B 16 湖 DB43 南 省 地 方 标 准 DB43/T 1 9 65—2020 赤子爱胜蚯蚓实验室饲养 与繁殖技术规程 Eisenia foetida Laboratory Feeding and Reproduction Technology Procedures 2020-12-29发布 湖南省市场监督管理局 2021-03-29实施 发 布 DB43/T 1965—2020 目 次 前言························································································································ Ⅲ 1 范围 ····················································································································· 1 2 规范性引用文件 ······································································································ 1 3 术语和定义 ············································································································ 1 3.1 生殖带 Clitellum ··························································································· 1 3.2 成蚓 Adult worm ····························································································· 1 3.3 蚓茧 Worm cocoon ··························································································· 1 4 环境要求 ··············································································································· 1 4.1 室温 ··············································································································· 1 4.2 光照 ··············································································································· 1 4.3 湿度 ··············································································································· 1 5 基质 ····················································································································· 2 6 饲养设备 ··············································································································· 2 7 饲料 ····················································································································· 2 8 饲养与繁殖方法 ······································································································ 2 8.1 成蚓的饲养 ······································································································ 2 8.2 繁殖方法 ········································································································· 2 9 观察记录 ··············································································································· 3 附录 A(资料性) 赤子爱胜蚯蚓图 ················································································ 4 附录 B(资料性) 赤子爱胜蚯蚓组织结构及截面图 ··························································· 5 I DB43/T 1965—2020 II DB43/T 1965—2020 前 言 本文件按照 GB/T 1.1—2020 给出的规则起草。 本文件某些内容可能涉及专利。本文件发布机构不承担识别这些专利的责任。 本文件由湖南省农业农村厅提出。 本文件由湖南省农业标准化技术委员会归口。 本文件起草单位:湖南省植物保护研究所,长沙艾格里生物科技有限公司。 本文件主要起草人:陈昂、刘勇、张德咏、张战泓、熊浩、陈武瑛、罗香文、刘志邦。 III DB43/T 1965—2020 IV DB43/T 1965—2020 赤子爱胜蚯蚓实验室饲养与繁殖技术规程 1 范围 本文件规定了赤子爱胜蚯蚓实验室饲养与繁殖技术中的赤子爱胜蚯蚓环境要求、基质、饲养设备、 饲料、饲养与繁殖方法和观察记录。 本文件适用于湖南省试验用赤子爱胜蚯蚓实验室饲养与繁殖。 2 规范性引用文件 下列文件中的内容通过文中的规范性引用而构成本文件必不可少的条款。其中,注日期的引用文件, 仅该日期对应的版本适用于本文件;不注日期的引用文件,其最新版本(包括所有的修改单)适用于本 文件。 3 术语和定义 下列术语和定义适用于本文件。 3.1 生殖带 Clitellum 蚯蚓身体前端表皮上的一种腺体,为鞍状或环带状,通常可通过颜色与蚯蚓身体其他部分区分开。 3.2 成蚓 Adult worm 身体前端呈现出生殖带的蚯蚓。 3.3 蚓茧 Worm cocoon 蚓茧多为椭圆形,一般只有半粒绿豆大,壁由粘膜硬化而成,又薄又软。蚓茧埋藏在土中,在温度 适宜时,孵化成幼蚓。 4 环境要求 4.1 室温 以(18~22)℃为宜。 4.2 光照 黑暗环境条件。 4.3 湿度 环境的湿度维持在 50%~80%为宜。 1 DB43/T 1965—2020 5 基质 饲养时所用的基质为 pH 为 5.5~6.5 的泥炭鲜。将泥炭藓充分捏碎,加入去氯自来水,搅拌均匀 至捏团不流水、松开不散落的状态,并确保添加的水不含氯。不定期添加饲喂水,使基质湿度保持在 30%~40%。 6 饲养设备 饲养箱约 16L(长 0.4m,宽 0.2m,高 0.2m)的塑料整理箱,亦可用木质箱。切去箱盖中间区域(即 约 30%面积) ,保留箱盖边缘约 6 cm 左右,在切去的区域覆盖 80 目纱网后用溶胶固定,作为饲养箱。 7 饲料 为灭菌牛粪或浸泡后灭菌的紫花苜蓿颗粒。 8 饲养与繁殖方法 8.1 成蚓的饲养 在清洗好的饲养箱中放入占饲养箱体积约 1/3 的上述基质,放入 300 条左右体色红润、大小均一、 具生殖带、活跃的赤子爱胜蚯蚓的成蚓。第 2 天观察成蚓情况,若未发现蚯蚓逃逸或死亡,则饲喂饲料 (饲喂量约为 500g) ,此后每七天定期饲喂一次,饲喂前一天需检查饲料剩余情况,若剩余量超过饲喂 量的 1/3,则减少饲喂量。 8.2 繁殖方法 8.2.1 成蚓的分离 成蚓连续培养 4 周,将基质倒在干净的托盘中,将基质中全部的成蚓挑出,使成蚓上不带有基质。 将成蚓放入另外一个加了约 1/3 基质的饲养箱中(基质的成分与 5 中相同) ,挑出的成蚓继续按照步骤 8.1 中方法培养四周。四周后按照步骤 8.2.1 的方法继续分离。 8.2.2 蚓茧的培养 取 8.2.1 分离成蚓后剩下的基质,所述基质内含大量蚓茧。将基质倒回原饲养箱中继续培养。保持 基质湿润,无需饲喂。 8.2.3 蚓茧孵化期间的培养 蚓茧培养 1 周之后,开始有幼蚓孵化,每周定期饲喂一次饲料(饲料为灭菌牛粪或浸泡后灭菌的紫 花苜蓿颗粒)且随时观察,培养 4 周时,蚓茧基本上全部孵化为幼蚓,蚓茧孵化率可达到 90%以上。 8.2.4 幼蚓分离 培养 8 周左右,目测幼蚓身体直径约有 1 mm 时,将基质倒在干净的托盘中,挑出全部的幼蚓。将 幼蚓分为三箱,在每个装有约 1/3 基质的饲养箱(基质的成分与 5 相同)中放入约 300 条幼蚓。 2 DB43/T 1965—2020 8.2.5 幼蚓培养 在幼蚓分离的第二天对幼蚓继续仔细检查,若幼蚓正常(未发现蚯蚓逃逸或死亡) ,则饲喂饲料(饲 料为灭菌牛粪或浸泡后灭菌的紫花苜蓿颗粒) 。此后每周定期饲喂一次饲料;继续培养约 6 周左右,幼 蚓基本上已经成为具生殖带的成蚓。从蚓茧孵化到试验用成蚓,大约需要 14 周,培养的成蚓可用于试 验与繁殖。 9 观察记录 及时观察记录蚯蚓的饲养、分离情况,一个星期(可根据实际情况)记录环境温度、湿度等情况。 3 DB4
